Main Title

MOTS-c 20 mg — Mitochondrial-Derived Peptide for Metabolic Homeostasis Research

Full Description

MOTS-c (mitochondrial open reading frame of the 12S rRNA type-c) is a 16-amino-acid peptide encoded within the mitochondrial 12S rRNA gene rather than the nuclear genome. Identified in 2015, it is investigated in preclinical research as a mitochondrial-derived peptide (MDP) that regulates metabolic homeostasis, AMPK signaling, insulin sensitivity, and adaptive gene expression under metabolic stress in models of obesity, aging, and energy imbalance.

What It Is

  • Class: Mitochondrial-derived peptide (MDP / microprotein)

  • Concept: Encoded in mtDNA and studied as a mitochondria-to-nucleus signaling peptide that activates AMPK, supports glucose and lipid utilization, and helps coordinate cellular energy and stress responses.

  • Format: Research-grade synthetic peptide supplied as a lyophilized powder.

Proposed Mechanisms (Preclinical)

AMPK & One-Carbon Metabolism

  • Investigated for modulation of the folate–methionine cycle and de novo purine biosynthesis, leading to AICAR accumulation and AMPK activation.

  • AMPK activation is studied for downstream effects on glucose uptake, fatty-acid oxidation, and energy-sensing pathways in skeletal muscle models.

Mitochondria-to-Nucleus Signaling

  • Under metabolic or oxidative stress, MOTS-c is studied for translocation from mitochondria to the nucleus.

  • In the nucleus, it is investigated for regulation of antioxidant-response and adaptive gene-expression programs.

Muscle & Metabolic Regulation

  • Explored for direct interaction with casein kinase 2 (CK2) in skeletal muscle and effects on insulin signaling, GLUT4-related glucose handling, and mitochondrial bioenergetics.

Selected Research Highlights

  • Metabolic Homeostasis: Landmark rodent studies reported prevention of diet-induced obesity and age-associated insulin resistance, with improved glucose handling in skeletal muscle models.

  • AMPK Pathway Research: Extensively used to study AMPK-dependent metabolic switching, fatty-acid oxidation, and energy-sensor signaling.

  • Exercise & Physical Performance Models: Investigated as an exercise-responsive peptide associated with improved running capacity and muscle metabolic function across age groups in mice.

  • Mitochondrial Bioenergetics: Studied for effects on intrinsic mitochondrial efficiency, reduced ROS emission, and PGC-1α/AMPK-dependent muscle bioenergetic performance.

  • Liver & NASH Models: Explored for interaction with Bcl-2 and improvements in mitochondrial metabolic pathways in experimental nonalcoholic steatohepatitis.

  • Aging & Stress Adaptation: Circulating MOTS-c levels have been examined in relation to age and metabolic disease; nuclear translocation research focuses on adaptive stress-response gene programs.
